Cleared vs. Non-Cleared 3D Cell Culture Models

Visikol is the only contract research organization capable of imaging 3D cell culture models (e.g. organoids, spheroids, microtissues) in their entirety through using the patented Visikol HISTO-M technology. As shown below, the addition of high throughput tissue clearing to 3D cell culture models allows for a dramatic increase in the number of cells detected and enables complete characterization of these models for use in drug discovery. 

Current assays are limited to characterizing the periphery of these models which significantly biases results and can lead to misleading evaluations of the impact of compounds/ antibodies on 3D cell culture models. 

Below: 250 um diameter HepG2 spheroids were labeled with DAPI nuclear staining and then imaged with a CellInsight CX7 LZR high content confocal system in Nunc 96-well plates. 1 spheroid was cleared with Visikol HISTO-M and one was imaged in PBS. Tissue clearing allows for a dramatic improvement in image quality and increases the number of cells that can be detected 3-4X.
